Caszyme’s research published in EMBO reports journal. This study introduces Cas12l, a new type V RNA-guided nuclease recognizing a unique C-rich PAM (5′-CCY-3′). Unlike typical type V nucleases, Cas12l exhibits optimal activity between 37 and 52°C, with a preference for cutting supercoiled DNA. It also demonstrates collateral ssDNA and ssRNA cleavage upon target recognition, suggesting its potential as a versatile genome editing tool.
