Project aim – to improve quality of personal health care services by creating rapid, accurate and for everyday health care institutions‘ use adapted CRISPR nuclease based detection method for SARS-CoV-2 virus and other infectious diseases.
Project description. Covid-19 disease emerged in 2019 in China and spread throughout the World causing a worldwide pandemic. Many countries had difficulties fighting the infection and in order to effectively control the pandemic extensive testing is crucial. Currently the most accurate detection method of the disease is based on collecting the sample with a nasal swab, purifying virus RNA, performing reverse transcription and real-time PCR (RT-PCR). That requires special equipment. These tests are costly and time consuming. In order to expand testing capacity, it is necessary to develop novel cheaper and faster but at the same time as accurate tests. One of the possible ways to create such test is by using CRISPR nucleases for detection of virus RNA. These enzymes use short crRNA to detect genome of the virus, nicks single stranded DNA probe and produces secondary analytical signal. Besides that using saliva as a sample would simplify and speed up testing process since medical staff, tools for taking the samples and medias for transportation of the samples would no longer be necessary. Improved test should not require additional purification step and isothermal amplification methods would be used for amplification of virus nucleic acids. We hope that by combining these steps it would help to speed up the testing process, lower the cost of the test and it be useful in everyday practice of health care institutions.
This project includes several tasks, that combined together would help to achieve results more effectively. During the project samples from patients will be gathered and bio banked in order to speed up the test validation process. At the same time another part of the project focused on optimization of direct use of saliva samples and isothermal virus RNA amplification will be conducted. Besides that characterization of few novel Cas nucleases is planned. That will be done in order to select the one that would be the most suitable for SARS-CoV-2 detection. Lastly, all the parts of the test will be combined together, and optimized protocol will be prepared. We hope that research conducted during this project not only allows to develop and validate a more convenient test for Covid-19 diagnostic but also builds a foundation for universal, easily programmable platform of diagnostics, that would enable detection of any pathogen of interest in the future.

Patobulintas testas turėtų būti be papildomos gryninimo stadijos, o viruso nukleorūgštys padauginamos izoterminės amplifikacijos būdu. Tikimės, kad šių žingsnių apjungimas padės pagreitinti ir atpiginti testą, kuris būtų tinkamas kasdienei gydymo įstaigų praktikai.
Šis projektas sudarytas iš kelių veiklų, kurių visuma padės efektyviau pasiekti užsibrėžtą rezultatą. Projekto metu rinksime ir biobankuosime pacientų mėginius, kad būtų galima greitai validuoti sukurtą testą. Lygiagrečiai bus vykdoma kita projekto dalis, skirta seilių kaip mėginio panaudojimo tiesioginiam tyrimui bei izoterminio viruso RNR padauginimo stadijų optimizavimui. Be to, planuojame charakterizuoti keletą naujų Cas nukleazių ir išrinksime tą, kuri geriausiai tinka SARS-CoV-2 aptikimui.
